In September 2018, after 57 years of operating in Canada, Dunkin' Donuts ceased business there when it refused to renew its franchise license to the few remaining stores left. All remaining Canadian locations were closed or rebranded as independent businesses in late 2018, ending the presence of Dunkin' Donuts in the country. === Otto Mostyn === Otto Mostyn (Roger Barclay) is Henry's godfather, and a fellow classmate of Charles Hanani from Oxford alongside Norton. A sinister, calculating businessman, Otto takes interest in Harper due to her cunning and ruthlessness, and agrees to provide seed funding to LeviathanAlpha, the hedge fund she is starting with Petra Koenig. Otto, Norton, and Aurore are also in a secret cabal to advance their interests via the government. Petra calls Otto after learning that Harper committed insider trading by acting on confidential information she overheard to short Pierpoint. Otto summons Harper to a meeting, where instead of reprimanding her, he tells her he wants her to succeed him in running his investment fund. Harper and Petra ultimately decide to call off the short and end their partnership with Otto to keep their fund above-board, but Harper later teams up with Otto to start her own fund dedicated to shorting corrupt companies over whom she and Otto would have leverage. In series 4, Otto becomes a member of the House of Lords and hopes to obtain a peerage. He puts increasingly strict constraints on Harper's trades at his fund, not wanting her to jeopardize his political connections; she eventually cuts ties with Otto. == Mechanism of action == Aprepitant is classified as an NK1 antagonist because it blocks signals given off by NK1 receptors. This, therefore, decreases the likelihood of vomiting in patients. NK1 is a G protein-coupled receptor located in the central and peripheral nervous system. This receptor has a dominant ligand known as Substance P (SP). SP is a neuropeptide, composed of 11 amino acids, which sends impulses and messages from the brain. It is found in high concentrations in the vomiting center of the brain, and, when activated, it results in a vomiting reflex. In addition to this it also plays a key part in the transmission of pain impulses from the peripheral receptors to the central nervous system. Aprepitant has been shown to inhibit both the acute and delayed emesis induced by cytotoxic chemotherapeutic drugs by blocking substance P landing on receptors in the brain's neurons. Positron emission tomography (PET) studies, have demonstrated that aprepitant can cross the blood–brain barrier and bind to NK1 receptors in the human brain. It has also been shown to increase the activity of the 5-HT3 receptor antagonist ondansetron and the corticosteroid dexamethasone, which are also used to prevent nausea and vomiting caused by chemotherapy. In addition to its activity as an NK1 receptor antagonist, aprepitant has also been identified as a microtubule-targeting agent (MTA). A nanoDSF-based screen with follow-up assays showed that aprepitant binds tubulin and completely inhibits microtubule polymerization in vitro, placing it among MTAs widely used in anticancer therapy.
=== Neoantigens === Many tumors express mutations. These mutations potentially create new targetable antigens (neoantigens) for use in T-cell immunotherapy. The presence of CD8+ T cells in cancer lesions, as identified using RNA sequencing data, is higher in tumors with a high mutational burden. The level of transcripts associated with the cytolytic activity of natural killer cells and T cells positively correlates with mutational load in many human tumors. In non–small cell lung cancer patients treated with lambrolizumab, mutational load shows a strong correlation with clinical response. In melanoma patients treated with ipilimumab, the long-term benefit is also associated with a higher mutational load, although less significantly. The predicted MHC binding neoantigens in patients with a long-term clinical benefit were enriched for a series of tetrapeptide motifs that were not found in tumors of patients with no or minimal clinical benefit.
